Fluorescence Microscope Dichroic Filters
Tuniu Optics’ excitation, emission and dichroic filters are custom-designed for fluorescence imaging, compatible with commonly used fluorescent dyes: DAPI, ATTO465, Alexa Fluor® 488, FITC, and TRITC. Optimized for central wavelength and spectral bandwidth, our fluorescence microscope filters feature high transmittance, deep blocking depth and steep spectral edges. They greatly suppress inter-channel crosstalk and deliver excellent signal-to-noise ratio for microscopic fluorescence imaging.
A dichroic mirror is designed to reflect light below a specified cutoff wavelength while transmitting light of longer wavelengths. In fluorescence microscopes, it directs light of the required wavelength range toward the sample and image plane. When placed at a 45° angle to the incident light in the optical path, the dichroic mirror reflects excitation light toward the sample and microscope objective, while the fluorescence emitted from the sample passes through the mirror and enters the detection system.
Though dichroic mirrors play an indispensable role in fluorescence microscopy, their performance in blocking unwanted light is not perfect. Typically, they reflect approximately 90% of light below the cutoff wavelength and transmit around 90% of light above it. As a result, part of the residual excitation light may transmit through the mirror together with the longer-wavelength fluorescence from the sample. An emission filter is therefore required to block such unwanted light and prevent it from reaching the detection system.
